Overview

Recombinant Human Tumor Necrosis Factor-alpha (TNF-α) is a key mediator of inflammation and immune regulation, produced primarily by activated macrophages, monocytes, neutrophils, T lymphocytes, and natural killer cells. It functions in host defense, tumor regression, and modulation of immune responses. TNF-α induces apoptosis in sensitive cell types, promotes cytokine production, and enhances leukocyte recruitment to inflamed tissues. The recombinant protein expressed in CHO cells retains native folding and bioactivity, making it ideal for in vitro research into inflammation, cytotoxicity, and signal transduction pathways.

Key Features and Benefits

  • Biologically Active: Exhibits ED₅₀ ≤ 0.02 ng/mL based on murine L929 cytolysis assay with Actinomycin D.
  • Low Endotoxin Level: < 1.0 EU/µg as per LAL testing—suitable for sensitive biological assays.
  • High Purity: > 95% confirmed by SDS-PAGE for reproducibility in critical experiments.
  • Validated Cytotoxic Activity: Proven to induce apoptosis in susceptible target cells in vitro.
  • Stable Lyophilized Form: Ensures long-term storage and consistent performance.

Reconstitution Instructions

To ensure optimal bioactivity and stability:

  1. Centrifuge the Vial: Briefly spin to collect the powder at the bottom.
  2. Reconstitute: Add sterile distilled water to a final concentration ≥ 0.1 mg/mL.
  3. Mix Gently: Swirl to dissolve; avoid vigorous shaking or vortexing.
  4. Dilute for Use: Further dilute in the desired buffer containing carrier protein.
  5. Storage: Store reconstituted protein at 2–8°C (≤ 1 month) or –20°C (≤ 6 months).

Quality Control (QC)

  • Purity: Verified by SDS-PAGE
  • Endotoxin: Assessed using Limulus Amebocyte Lysate (LAL) method
  • Bioactivity: Confirmed by cytolysis assay with murine L929 cells in the presence of Actinomycin D
