Overview
Recombinant Human Neurotrophin-3 (NTF3, also known as NT-3) is a critical member of the neurotrophin family that regulates survival, differentiation, and maintenance of specific neuronal populations. It is naturally expressed in central nervous system neurons and muscle tissue, with reduced expression in denervated muscles. NT-3 shares structural domains with NGF and BDNF, showing additive biological effects with BDNF in certain contexts. It has been shown to prevent apoptosis of cultured embryonic rat spinal motor neurons at picomolar concentrations and to promote corticospinal tract sprouting during development and after spinal cord injury.
The biological activities of NT-3 are mediated through Trk family tyrosine kinase receptors; however, NT-3 only binds weakly to the Trk receptor that serves as the high-affinity receptor for NGF.
Key Features and Benefits
- Validated Bioactivity: Demonstrates ED₅₀ of 10–50 ng/mL in choline acetyltransferase induction assay in primary septal neurons.
- High Purity, Low Endotoxin: >95% purity by SDS-PAGE; endotoxin level < 1.0 EU/µg for sensitive applications.
- Neuroprotective Role: Supports neuronal survival and axonal regeneration in both developmental and injury models.
- Broad Research Utility: Suitable for studies in neurobiology, CNS repair, and comparative neurotrophin signaling.
- Stable Format: Lyophilized powder allows long-term storage and convenient reconstitution.
Reconstitution Instructions
To prepare for use, follow these procedures carefully:
- Centrifuge the Vial: Spin briefly to ensure powder is collected at the bottom.
- Reconstitute: Add sterile distilled water to achieve a final concentration of ≥ 0.1 mg/mL.
- Dissolution: Swirl gently until fully dissolved. Do not vortex.
- Dilution and Application: Dilute with desired buffer or culture medium for immediate use.
- Storage: Store reconstituted solution at 2–8°C for up to 1 month, or –20°C for up to 6 months with carrier protein. Avoid repeated freeze/thaw cycles.
Quality Control (QC)
- Purity confirmed by SDS-PAGE (>95%)
- Endotoxin level measured by LAL assay (<1.0 EU/µg)
- Bioactivity verified in choline acetyltransferase induction assay in rat primary septal neurons
