Overview

Recombinant Human Interleukin-1 Beta (IL1B) is a potent immunomodulatory cytokine involved in the regulation of inflammatory responses. It is primarily secreted by monocytes and macrophages and plays a vital role in host defense by promoting fever, activating endothelial cells, and stimulating the production of acute-phase proteins.

IL1B functions as a monomer and must be enzymatically cleaved from its precursor form to become biologically active. It shares a receptor with IL1A, and both isoforms elicit similar signaling effects. IL1B has been shown to stimulate the release of prostaglandins and collagenases from synovial cells, induce thymocyte proliferation, B-cell maturation, and fibroblast growth factor activity, making it essential for studying inflammatory and autoimmune pathways.

Key Features and Benefits

  • Proven Biological Activity: Demonstrates ED₅₀ < 1 ng/mL based on inhibition of prostate carcinoma cell proliferation.
  • High Purity, Low Endotoxin: Purity > 95% verified by SDS-PAGE; endotoxin content < 1.0 EU/µg, suitable for sensitive cell assays.
  • Inflammation Research Tool: Facilitates exploration of cytokine-mediated responses in innate immunity and autoimmunity.
  • Receptor Binding Fidelity: Binds IL1 receptor to mimic native IL1B signaling without loss of activity.
  • Stable Format: Supplied as a lyophilized powder for extended shelf life and convenient reconstitution.

Reconstitution Instructions

To prepare for use, follow these procedures carefully:

  • Centrifuge the Vial: Briefly spin the vial to collect the contents.
  • Reconstitute: Add sterile distilled water to achieve a concentration ≥ 0.1 mg/mL.
  • Dissolution: Swirl gently to dissolve the protein completely. Do not vortex.
  • Dilution and Application: Further dilute using the desired buffer or medium for experimental use.
  • Storage: After reconstitution, store at 2–8°C for up to 1 month, or at –20°C for up to 6 months (with carrier protein). Avoid freeze/thaw cycles.

Quality Control (QC)

  • SDS-PAGE confirmed purity > 95%
  • Endotoxin level verified by LAL assay (< 1.0 EU/µg)
  • Biological activity validated using carcinoma cell inhibition assay
