Culture & Handling Guidelines
Recommended Culture Conditions
- Culture Vessel: Use suspension culture flasks or low-adherence plates to maintain cell viability.
- Growth Medium: PriGrow X Series Medium (TM4124) supplemented with:
- 1% Penicillin/Streptomycin Solution (G255)
- 50 ng/mL Recombinant Human IL-4 (Z100545)
- 50 ng/mL Recombinant Human GM-CSF (Z100275)
- Incubation Conditions: Maintain at 37°C in a humidified incubator with 5% CO₂.
- Seeding Density: 2×104 – 5×104 cells/cm2
- Cell Proliferation: Human CD56+ NK Cells do not proliferate in vitro and should be used within the recommended timeframe for optimal viability.
Thawing Protocol
- Remove the cryovial from liquid nitrogen storage and immediately transfer it to a 37°C water bath.
- Thaw rapidly (within 2 minutes) while gently agitating the vial. Keep the cap above the water level to avoid contamination.
- Spray the vial exterior with 70% ethanol, then transfer it into a biosafety cabinet.
- Pipette the cell suspension into a 15 mL sterile conical tube containing 5 mL pre-warmed complete growth medium.
- Centrifuge at 400 × g for 10 minutes to pellet the cells.
- Carefully aspirate the supernatant without disturbing the cell pellet.
- Resuspend the cell pellet in fresh pre-warmed complete growth medium and transfer to a suitable culture vessel.
- Incubate under recommended conditions for at least 4 hours before use to allow for cell recovery.
Subculturing Guidelines
- Human CD56+ NK Cells do not proliferate under standard culture conditions.
- It is recommended to use freshly thawed cells without extended passaging.
- If expansion is required, consider co-culture with feeder cells or cytokine stimulation protocols.
Cryopreservation Guidelines
- Cryopreservation Medium: Not recommended for re-freezing.
- Storage Conditions: Cells must be stored in the vapor phase of liquid nitrogen or below -130°C for long-term preservation.
