Overview

Neuregulin-4 (NRG4) is a member of the neuregulin family of EGF-like ligands that activate ErbB receptor tyrosine kinases, particularly ErbB4. These signaling events regulate cell proliferation, differentiation, and metabolic homeostasis, with implications in neural development, cardiac function, and energy metabolism. NRG4’s selective interaction with ErbB4 enables targeted activation of downstream pathways, making it a valuable molecular tool for research in metabolic disorders, neurobiology, and tissue regeneration. This recombinant NRG4 is expressed in E. coli, purified to high homogeneity, and retains robust biological activity in cell-based assays.

Key Features and Benefits

  • High Potency: ED₅₀ < 0.05 ng/mL in Raji cell proliferation assay.
  • Defined Structure: Recombinant protein expressed in E. coli, ~7 kDa, representing the biologically active domain.
  • High Purity & Low Endotoxin: >95% purity, <1.0 EU/µg endotoxin, suitable for sensitive biological systems.
  • Reproducible Performance: Lot-to-lot consistency confirmed through rigorous QC testing.
  • Versatile Application: Applicable to studies in ErbB4 signaling, metabolic regulation, neural development, and cancer biology.

Reconstitution Instructions

To ensure optimal protein quality and activity:

  1. Centrifuge the Vial: Briefly spin to collect all lyophilized material at the bottom.
  2. Reconstitute: Add sterile distilled water to achieve a final concentration of not less than 0.1 mg/mL.
  3. Mixing: Gently swirl or tap the vial to fully dissolve the protein. Do not vortex.
  4. Dilution and Application: After complete reconstitution, dilute the solution with appropriate buffers or media containing carrier protein, as required for your experiment.
  5. Storage: Store reconstituted solution at 2–8°C for up to 1 month, or –20°C for up to 6 months (preferably in aliquots). Avoid repeated freeze/thaw cycles to maintain stability and bioactivity.

Quality Control (QC)

  • Purity confirmed by SDS-PAGE
  • Endotoxin measured by LAL assay
  • Biological activity validated in Raji cell proliferation assay
