Overview
Erythropoietin (EPO) is a glycoprotein hormone essential for the regulation of erythropoiesis. In the human body, it is primarily synthesized and secreted by the kidney’s tubular and juxtatubular capillary endothelial and interstitial cells, with a smaller proportion produced by hepatocytes and Kupffer cells in the liver. Approximately 40% of its molecular mass results from glycosylation, which plays a critical role in its pharmacokinetic properties. Non-glycosylated forms are rapidly cleared in vivo, whereas the glycosylated form maintains biological activity and stability. This recombinant human EPO is expressed in CHO cells, ensuring proper glycosylation and folding, and is ideal for in vitro studies focused on erythropoiesis, hematopoietic cell biology, and anemia-related research.
Key Features and Benefits
- Potent Biological Activity: Stimulates TF-1 cell proliferation with ED₅₀ < 0.2 ng/mL.
- Glycosylated for Stability: CHO cell expression ensures natural glycosylation, prolonging biological half-life.
- High Purity & Low Endotoxin: >95% purity (SDS-PAGE) and <1.0 EU/µg endotoxin, suitable for sensitive cell assays.
- Validated Reproducibility: Lot-to-lot consistency confirmed through QC testing.
- Versatile Application: Suitable for research in hematopoiesis, anemia models, and erythroid differentiation studies.
Reconstitution Instructions
To ensure optimal protein quality and activity:
- Centrifuge the Vial: Briefly spin to collect lyophilized material at the bottom.
- Reconstitute: Add sterile distilled water to achieve a final concentration of not less than 0.1 mg/mL.
- Mixing: Gently swirl or tap the vial to dissolve; do not vortex.
- Dilution and Application: Further dilute with appropriate buffer containing carrier protein as required.
- Storage:
- Store reconstituted solution at 2–8°C for up to 1 month, or –20°C for up to 6 months.
- Avoid repeated freeze/thaw cycles to maintain protein stability and bioactivity.
Quality Control (QC)
- Purity confirmed by SDS-PAGE
- Endotoxin measured by LAL assay
- Bioactivity validated in TF-1 cell proliferation assay
