Overview

Recombinant Human Interleukin-33 (IL-33) is a member of the IL-1 cytokine family and functions as a potent inducer of Th2-type immune responses. IL-33 signals through the ST2/IL-1RAcP receptor complex, leading to NF-κB and MAPK (ERK1/2, p38, JNK) activation in mast cells and other immune subsets. It is expressed in a variety of cell types including epithelial cells and fibroblasts, and facilitates enhanced production of IL-5 and IL-13 from Th2 cells. IL-33 also acts as a chemoattractant, contributing to the recruitment of Th2 lymphocytes during allergic and inflammatory responses.

Key Features and Benefits

  • Biologically Active: Exhibits high potency with an ED₅₀ of ~0.03 ng/mL based on D10S cell proliferation assay.
  • Endotoxin Controlled: < 1.0 EU/µg as per LAL testing—appropriate for sensitive systems.
  • High Purity: > 95% verified by SDS-PAGE analysis ensures reproducibility in critical assays.
  • Validated Immune Modulation: Activates NF-κB and MAPK signaling; enhances Th2 cytokine responses in vitro.
  • Stable Lyophilized Form: Enables long-term storage and consistent performance.

Reconstitution Instructions

To ensure optimal bioactivity and stability:

  1. Centrifuge the Vial: Briefly spin down to collect the powder at the bottom.
  2. Reconstitute: Add sterile distilled water to a final concentration ≥ 0.1 mg/mL.
  3. Mix Gently: Swirl to dissolve; avoid vigorous shaking or vortexing.
  4. Dilute for Use: Further dilute in desired buffer containing carrier protein.
  5. Storage: Store reconstituted protein at 2–8°C (≤ 1 month) or –20°C (≤ 6 months).

Quality Control (QC)

  • Purity: Verified by SDS-PAGE
  • Endotoxin: Assessed using Limulus Amebocyte Lysate (LAL) method
  • Bioactivity: Confirmed in cell-based proliferation assay with D10S murine cells
